Luke Farrell was selected by the Jacksonville Jaguars in the fifth round, 145th overall, of the 2021 NFL Draft. He signed a four-year rookie contract with the team on May 20, 2021. Farrell appeared in all 17 games for the Jaguars during the 2021 season, recording his first catch in a Week 2 loss to the Denver Broncos, 13–23. He ended the season with seven receptions for 56 yards and no touchdowns. His longest catch was 21 yards, achieved during a Week 6 23–20 win over the Miami Dolphins.
